Influence of Diuretic on Excretion of ^(203)Hg Chlormerodrin

Abstract
In this experiment the animals used were adult mongral female dogs with body weight of 1014 kgs. The dog was first anesthetized with pentobarbital and hydrated with 0.9 o Nacl. Then, after an injection of 203 Hg chlormerodrn first and then additional injection of mercuro-phylline, ethacrinic acid and respectively, the following factors¢¥ were studied and compared: The quanitity of ~03Hg chlormerodrin in the excreted urine RPF GFR, urinary volume, -blood content of =03Hg chlormerodrin and the quantity of zosHg chlormerodrin in the kidney tissue.
The total of 35 experimental dogs were divided -into two groups, A and B.
Group A: In this group the dogs were injected through femoral vein with ~03Hg chlormerodrin and then an additional injection of merc;urophyilline, ethacrgnic acid and furosemide respectively, and then the urinary volume, sodium and potassium content in the excreted urine;, the radioact activity of ~3Hg chlormerodrin in ¢¥blood, in the excreted quantity of ~3Hg chlomerodrin the urine and the accumulated quantity of the same in the kidney tissue were studied and compared.
Group B: In this group the dogs were injected through femoral vein with ~03Hg chlormerodrin as above, but the ethacrinic acid and fiirosemide were injected through one of the renal artery respectively, and then the excreted volume of the urine, the radioactivity of ~03Hg inblood the quantity of =03Hg chlormerodrin in the urine,, RPF, GFR and the accum~ilated quantity of =03Hg clormerodrin in the kidney tissue of each kidney were studied and, compared respectively. From the above stuGroup A: 1) The urinary volume was not increased in the group of dogs with injection of ¢¥sosHg chlrmerodrin only and =03Hg chlormero-drin plus mercurophilline, but markedly increased in the dogs with =aHg chlormerodrin plus ethairrnic acid and euremide anc~ parti-cularly more marked in the group with furosemide.
5) The excreted quantity of 203Hg chlormerin in the urine were increased in the dogs with mercurophilline and those with chloromerodrin plus ethacrinic acid, but decreased in the dogs with chlormeorodrin only and chlomerodrin plus furosemide.
6) The accumulated quantity of ~03Hg chlormerodrin in the kidney tissue was least in the group with mercurophilline and about the same among the others.
Group B. The injection site was selected at the left renal artery.
1) The urinary volume was markedly increased in the left kidney of the dogs with ethacrinic acid and furosemaide injection group and particularly more marked in the furosemide inject-ion group.
2) There were no significant differences of R. P. F and G. F. R in each kidney.
3) The excreted quantity of 203Hg was very markedly increased in the group with ethrarinic acid injection.
4) The accumulated quantity of =03Hg in the kidney tissue showed no significant differences among the group.
From this study the ethacrynic acid seems to increase very markedly the excretion of the =03Hg chlormerodrin in the urine. but the excretion of the 03Hg chlormerodrion does not seem to affect the urinary volume, R. P. F. and G. F. R very much.
It seems that the ethacrynic acid and mercurophylline compete each other for the combination with protein bound sulfhydryl of the cell iu vivo.
